On [Illumina](faq_NGStechnologies) type sequencers, RNA hast to be converted to cDNA first. Depending on the size of the RNA, a fractionation step is needed to reduce the size of the cDNA to be compatible with the cluster formation (20 to 1000bp).
On an Oxford Nanopre sequencer, both cDNA and also RNA can be directly sequenced if the RNA is available in a sufficient amounts.
When sequencing RNA on oxford nanopore modifications bases can be detected. The detection of RNA modifications is still under full development.
